What Is Anandamide?

The entrance is silky and exudes a thick milky fragrance, which instantly feels full of happiness. Yes, this is the rich chocolate brought to us.

Luckily, I have this opportunity to introduce you one "happiness molecule" that has a great relationship with chocolate - Anandamide(Sanskrit means "joy, bliss, delight")

Anandamide (also called N-Arachidonoylethanolamine or AEA), a naturally endogenous cannabinoid neurotransmitter, found in the mammalian brain, also present in food like Cocoa powder, Chocolate, Truffles.

Mentioned endogenous cannabinoid neurotransmitter, comfortably in your mind is cannabidiol (CBD), so what relations between them?

Cannabidiol and Anandamide

Cannabidiol (CBD) is a phytocannabinoid found in 1940. It is one of the famous compounds in cannabis plants.

2-Arachidonoylglycerol (2-AG), first discovered maternal bovine in 1994, is another one endocannabinoid, a central endogenous agonist of the CB1 receptor.

Anandamide was discovered in the brain of pigs in 1992 and is the second recognized endogenous ligand for cannabis found after 2-AG (2-Arachidonoylglycerol).

Many studies suggest that these endocannabinoids play essential roles in the regulation of mood, memory, appetite, and pain perception.

Name
2-Arachidonoylglycerol
Anandamide
Other names
2-AG
Arachidonoylethanolamide; AEA
Commonality
Both endogenous ligands of cannabinoid (CB) receptors;
Both arachidonic acid-containing lipid molecules
Difference
2-AG is a full agonist of the central receptor CB1 as well as the peripheral receptor CB2, Bubutanandamide is a partial agonist of both receptors;
Endogenous 2-AG level is higher than anandamide level in animal tissues;
2-AG is a significant product of the phospholipase Cβ - diacylglycerol lipase pathway, anandamide is a minor;
2-AG potentiates GABAA receptor, whereas anandamide is one of the vanilloid receptor, TRPV1, agonists

Biosynthesis and degradation of Anandamide

The synthesis of AEA requires Ca2+ to participate in two steps: first, phosphatidylethanolamine produces N-arachidonoyl-phosphatidylethanolamine (NArPE) catalyzed by Ca2+-dependent N-acyltransferase; then NArPE is in N-acyl AEA, and phosphatidic acid produced by phosphatidylethanolamine selective phospholipase D (NArPE-PLD).

Cellular reuptake and enzymatic degradation mechanisms inactivate AEA. It is diffused into cells by simple diffusion or diffusion under a special transporter and then hydrolyzed by fatty acid amide hydrolase (FAAH) to form arachidonic acid and ethanolamine to degrade.

Health Benefits of Anandamide

Anandamide has a wide-ranging effect on the body because it interacts with the endogenous cannabinoid system. Among many other functions, Anandamide plays a role in reward, mood, appetite, pain relief and reproduction.

Vasodilation–Anandamide can play an essential role in regulating the physiological functions of the cardiovascular system through neuromodulation mechanisms or directly acting on endothelial cells, vascular smooth muscle, and myocardium.

Neurogenesis–Anandamide can be a powerful chemical associated with neurogenesis especially in the elderly. Dr. Gary Wenk, a famous researcher of neuroscience, has insights into the effects of anandamide and the interactions with the brain.

Anti-Depressive–Anandamide enhances the mood and quality of life besides cognitive functions. Researches on rat brain models showed anandamide has a strong anti-depressive effect on the brain, which can improve a better mood.

Anandamide affects on many diseases

Schizophrenia
Anandamide in patients with acute schizophrenia was approximately three times higher than that in healthy subjects (7.79±0.50 / 2.58±0.28 pmol/ml). After treatment, the content of arachidonic acid ethanolamine in the control group decreased to near normal group (3.88±0.72 pmol/ml). In the early stages of mental illness, Anandamide also showed an up-regulation that the cannabis system has a protective effect on the first mental illness.

Patient Number
Anandamide levels in the blood during the acute phase of the schizophrenic disease(pmol/ml)
Anandamide levels in the blood during remission of schizophrenic disease (pmol/ml)

Changes of Anandamide content in plasma of people with a mental health condition

Pregnancy and abortion
The content of arachidonic acid ethanolamine in the blood of normal pregnant women has a relatively consistent pattern.

The first trimester> the second trimester =the third trimester < the term nonlaboring (0.89±0.14nM 0.44±0.12nM 0.44±0.11nM 0.68±0.09nM )

The term laboring women (2.5±0.22nM) had the largest Anandamide content in the blood.

This pattern of change has a protective effect on the successful completion of the pregnancy process. There is evidence that maintaining high Anandamide is detrimental to blastocyst production; this may be a contributing factor to abortion, fetal growth restriction, and preterm birth. It is not clear how the cannabis system regulates and manages these complex processes.

Anorexia nervosa and overeating disease

Since Anandamide can stimulate food absorption, the concentration of arachidonic acid amine in the plasma of patients with anorexia nervosa and overeating increased. It may be that the patient counteracts the restriction (constraints) by increasing the driving force of eating food. An adaptive response made by the influence.

Obesity and diabetes

Ordinary people with less fat in Anandamide in obese patients are about 35% higher on average; obesity is one of the leading causes of type 2 diabetes. The experimental results show that the peripheral cannabis system up-regulated in obese people. The plasma content of arachidonic acid in diabetic patients is also significantly higher than that of ordinary people. Many studies have shown that the peripheral cannabis system activated in type 2 diabetes.

More and more studies have shown that Cannabinoid Anandamide has undergone significant changes in the development of many diseases (such as depression, multiple sclerosis, and Huntington's disease, etc.) and may be involved in the process of these diseases.

The change of Anandamide in human body fluid may be the leading indicator for disease prevention, diagnosis, and treatment.
